Abstract The present study was conducted at the Genetics laboratory and Experimental Farm, Department of Horticulture Fac. Agric., EL-Minia University, to obtain some Bradyrhizobium genotypes (Trans formants) specific on mungbean. The response of some mungbean cultivars to inoculation with these transformants compared to inoculation with wild type was investigated in the open field of the Experimental Farm laboratory experiments: DNA was extracted :trom Rhizobium leguminosarum, which was resistant to 400µglml chloramphenicol and used as a donor in transformation, while the recipient was Bradyrhizobium spp sensitive to 50_glml of chloramphenicol antibiotic. After transformation, experimental results showed that the frequencies of transformants at IOO_glml of antibiotic were 63xl0-5%. Testing the transformants at a high rate of antibiotic (300_ glml Ch.), two transformants showed a full growth on this rate of antibiotic and these two transformants were used in the field experiment. Field experiments: Two field experiments were carried out during the two successive seasons; 1997 and 1998 at the Experimental Farm, Department of Horticulture. Fac. Agric., Minia University to study the response of some mungbean cultivars (Imported I, imported 2, Kawmy, Giza 1 and V2010) to inoculation with some Bradyrhizobium transformants. The wild type and transformants isolated from Bradyrhizobium, used in this study were Bradyrhizobium spp. trans formant (TI), Bradyrhizobium spp trans formant (T2), Bradyrhizobium spp wild type (Wt).
