Abstract

This study was undertaken to establish a model of liver fibrosis and to study the effect of losartan and/or propolis on prevention and reversion of liver fibrosis. On evaluation of the distribution of the fibrous tissue, it was found that the liver architecture became totally distorted after CCl4 injection for 8 ws. Two weeks after stopping CCl4 injection, the septa were less in thickness and the pseudolobules were larger in size. On evaluation of the activated HSCs/myofibroblasts, ?-SMA positive cells appeared in the sinusoidal wall after CCl4 injection and after stopping the injection, the expression of ?-SMA was markedly declined. By image analysis, the area occupied by collagen fibers was progressively increased with increase the period of CCl4 administration. It was found that losartan improved the pathological changes and reduced the elevated liver enzymes induced by CCl4. It reduced the amount of fibrous tissue, fibrosis area (detected by image analyzer) and fibrosis grade induced by CCl4. Losartan also reduced the ?-SMA expression induced by CCl4. losartan was effective when it was given either with or after the beginning of the injection. It decreased the fibrosis area but its effect was not significant when given after stopping the injection. It was found that propolis improved the pathological changes and decreased the elevated liver enzymes induced by CCl4 injection and its effect was more stronger than that of losartan. Propolis reduced the amount of fibrous tissue, fibrosis area and fibrosis grade induced by CCl4 injection. propolis was effective when given with or after 1w from the beginning of the injection and not effective when given after stopping the injection. It was found that combination of both losartan and propolis improved the CCl4-induced pathological changes and significantly reduced the elevated liver enzymes induced by CCl4 injection. Combination of both losartan and propolis markedly reduced the amount of fibrous tissue, fibrosis area and fibrosis grade induced by CCl4 injection. The combination had a greater effect on reducing the elevated liver enzymes, fibrosis area and fibrosis grade than the effect of each drug alone. It was found that combination of both drugs was more effective in reversing the liver fibrosis as the liver architecture was not distored. Combination of both drugs significantly decreased the fibrosis area and grade of liver fibrosis induced by CCl4.
