Abstract

This study evaluated the synergistic effects of Portulaca oleracea L. (purslane) seed oil methanolic extract (PSO) and doxorubicin (DOX) on liver and kidney function, oxidative stress, apoptosis, and immunomodulation in an Ehrlich Ascites Carcinoma (EAC) mice model. Eight groups of mice received different treatments: control, PSO, DOX, PSO+DOX, EAC, EAC+PSO, EAC+DOX, and EAC+PSO+DOX.
Results showed that in the EAC group, liver and kidney markers, AFP tumor marker, oxidative stress, and cell proliferation increased, while antioxidants, apoptosis, and live cells decreased compared to control. Immune checkpoints PD-L1 and TGF-β were upregulated. Liver histology showed damage, necrosis, inflammation, and fibrosis.
In groups treated with PSO alone or combined with DOX, liver and kidney markers improved, AFP decreased, oxidative stress reduced, antioxidants increased, apoptosis increased, live cells increased, PD-L1/TGF-β decreased, and proliferation markers decreased compared to EAC. Liver histology showed reduced damage.
Conclusion: PSO, especially with DOX, may protect against EAC damage, improve liver function, and modulate immunity. It could be a potential therapeutic option for liver diseases and cancer.
Recommendation: Further studies and clinical trials are suggested to validate purslane’s therapeutic applications.
