Abstract

Summary & Conclusion
Cancer is considered as one of the leading causes of death worldwide. Among different types of cancer, lung cancer and breast cancer are very common with high death rates. Early detection and treatment of cancer are essential for better health management and improved survival. There is an alarming increase in drug resistance against cancer treatments, and new drugs are urgently needed to surmount this issue. Cryptolepine and neocryptolepine are isolated from Cryptolepis sanguinolenta’s roots. Recently, it was shown that cryptolepine, neocryptolepine, and their derivatives have a promising anti-cancer activity. Here, 11- (1,4-Bisaminopropylpiperazinyl) neocryptolepine (a new derivative of neocryptolepine) was synthesized and its cytotoxic activity was assessed on breast and lung carcinoma cell lines and the possible mechanism was examined. BAPPN was synthesized and characterized with FT-IR and NMR spectroscopy. Treatment of MCF-7 and A549 cells with an IC50 of 3.1 μg/ml for MCF-7 cells and 9.96 μg /mL for A549 cells was done to examine its cytotoxic activity. MTT assay was performed to confirm its cytotoxic activity.
Then, flow cytometry analysis was conducted to examine the apoptotic activity of BAPPN. Finally, ELISA Assay was done to measure the expression of different apoptotic proteins such as caspase-3 and p53, and proliferative proteins such as VEGF, ki-67, and PCNA in both MCF-7, and A549 cell lines to identify the cytotoxic mechanism of BAPPN. The results showed that BAPPN induces cytotoxic activity against both cancer cell lines (MCF-7 and
