Abstract

Objective: This work was conducted to investigate the cardioprotective
effect of grape seed extract and L-carnitine against doxorubicin-induced
cardiotoxicity in rats.
Methods:
A total of 60 male albino rats. were divided into 6 groups, group I: rats
served as the control group and received 0.5 ml/kg b.wt. of normal saline
0.9% NaCl solution for the whole experimental period (35 days), group
II: given grape seed extract (GSE) (100 mg/kg b.wt., daily) for the whole
experimental period, group III: given L-carnitine (L-CA) (150mg/kg
b.wt., twice per week) for the whole experimental period, group IV:
given doxorubicin (DOX) i.p. on day 28 of the experiment to induce
cardiotoxicity (10mg/kg b.wt.), group V: given combination of GSE
+DOX, group VI: given combination of L-CA +DOX. On days 29 and
35 of the experiment. the following parameters were measured: body
weight, heart weight, ECG, hematological, some serum biochemical
parameters, lipid profile (TC, TAG, HDL-c, LDL-c, VLDL-c), cardiac
injury markers (CK-MB, LDH, AST, cTnI, and NT-proBNP), oxidative
stress markers and antioxidant enzymes (GSH, CAT, TAC, TOC, MDA,
TNO, and LOOH), inflammatory markers and cytokine analysis (IL-1
β, TNF-α, MPO, and NF-κB), histopathological examination of heart
was also performed.
Results: The results showed that GSE and L-CA significantly
ameliorated the ECG abnormalities, the levels of serum cardiac markers,
oxidative stress markers and antioxidant enzymes, and inflammatory,
and cytokine analysis that deteriorated by DOX. Histopathological
examination of heart sections of rats pre-treated with GSE and L-CA
showed alleviation of histological degenerative changes caused by
DOX.
Conclusion: The results suggested that the GSE and L-CA induce
potent cardioprotective and antioxidant effects in DOX-induced
cardiotoxicity in rats.
