Abstract
This work has been carried out to investigate: 1- Spectrophotometric determination of glucose in pure form and in a synthetic embryos’ culture medium mixture through oxidation with TTC. Then, applying the successful procedure to the actual culture medium. Extraction of the product was performed and its characterization using elemental analysis, IR and NMR techniques was done. 2- Spectrophotometric determination of sodium pyruvate in pure form and in a synthetic embryos’ culture medium mixture through reaction with DNPH and the application to the actual human embryos’ culture medium. The solid product was isolated and characterized using elemental analysis, IR and NMR methods. 3- Spectrophotometric determination of the total amino acids (Gly and Trp) through reaction with ninhydrin in a binary and in a synthetic embryos’ culture medium mixture. The method was applied to estimate the total amino acids content in the actual human-embryos’ culture medium. Spectrophotometric determination of Trp through reaction with ferroin indicator at two different wavelengths. The work was also focused on the mechanism of reaction between Trp and ferroin. The successful procedure was applied to determine Trp in both synthetic and the actual human embryos’ culture medium.
