Abstract

High intake of dietary fructose is accused for responsibility for the development of the metabolic syndrome (MS). This work aimed to observe the beneficial effect of sitagliptin (Sita), pioglitazone (Pio), alpha lipoic acid (ALA), taurine (Tau) or their combination on fructose-induced MS. Ten groups of rats (n = 12) were fed on fructose enriched diet (FED) for 14 weeks followed by single i.p. injection of streptozotocin (STZ). One served as FED control while the remaining groups received sitagliptin (10 mg / kg. p.o.), pioglitazone (10 mg / kg. p.o.), Ü- lipoic acid (100 mg / kg .p.o.), Taurine (100 mg / kg. p.o.) or their combination, respectively, for the last 4 weeks. A fifth group was fed with on normal diet (normal control). At the end of the study body weight and systolic blood pressure (SBP) were measured, blood samples were collected for estimation of fasting blood glucose (FBG), fasting insulin (FINS), insulin resistance (IR), insulin sensitivity (IS), Ý- cell functions (BCF), triglycerides (TG), total cholesterol (TC), high density lipoproteins cholesterol (HDL - C), low density lipoproteins cholesterol (LDL - C), malondialdehyde (MDA) and uric acid as well as superoxide dismutase (SOD) and glutathione - S - transferase (GST) activities. Induction of MS resulted in significant increases in SBP, FBG, FINS, IR, TG, TC, LDL - C, MDA and uric acid. Meanwhile it resulted in a significant decrease in IS, BCF as well as SOD and GST activities. Treatment of rats with sitagliptin resulted in significant reduction in SBP, hyperglycemia, IR, TG, T. cholesterol, LDL - C, MDA and uric acid levels, as well as improvement in BCF and IS. Similarly, treatment of insulin resistant rats with pioglitazone showed similar results in addition to improvement in serum SOD activity
