Abstract

Silybum marianum seeds proved to have many therapeutic properties for the valuable components especially silymarin. Ethanolic extracts for silymarin and nano silymarin (100 mg/kg body weight) was measured through Ehrlich Ascites Carcinoma EAC bearing mice. The results of analytical chemical analysis and histological studies have been found as followed:
1- Liver enzymes recorded the highest value in positive EAC group as extremely high significant compared to negative control. Nano silymarin improved ALT by 51.24, and AST by 35.25% comparing to positive control. Silymarin improved ALT by 33.84, and AST by 29.7% compared to positive control.
2- Renal functions were increased significantly in urea, and very high significant with creatinine for EAC model. Comparing to positive control, nano particles improved creatinine by 27.94 and urea by 13.9%, while silymarin showed improvement by 26.98, and 9.58% respectively.
3- EAC mice increased the bilirubin compared to control by extremely high significant. Nano silymarin enhanced this record by 51.92, and silymarin did it by 45.51%.
4- Lipid profile increased dramatically in extremely high significant for EAC carcinoma mice model in triglycerides, cholesterol, risk factors Chl/HDL and LDL/HDL comparing to control. Nano silymarin improved total cholesterol by 25, triglycerides by 25.2, Chl/HDL by 50.25 and 50.39% for LDL/HDL. These enhancement for silymarin were 20.9, 17.9, 41.84, and 44.12% respectively.
5- In protein, albumin and A/G ratio showed extremely high significant decrease for EAC mice comparing to control. Nano silymarin showed immune system as the record of normal control, and that was better than that for silymarin treatment.
6- Tumar marker (α-fetoprotein) has very high significant in EAC group. This value was enhanced by nano particles by 43.72, and by silymarin by 41.85% compared to positive control.
7- LDH with extreme high significant was the result for EAC. This value was improved by 22.59, and 22.25% when using nano silymarin and silymarin respectively.
8- G6PD was also extremely high significant for positive control, and that was improved by nano silymarin in 44.55% and by silymarin by 17.92%.
9- The oxidative ensymes and reactive oxygen species experiments were measured. MDA was highly significant with EAC, and that was changed to 26.31 with nano silymarin and into 24.83 using silymarin. While, SOD showed reduction in non significant with EAC group, and that was enhanced by 50 with nano silymarin, and by 45.4 with silymarin respectively. CAT enzyme showed very high significant reduction with EAC group, and that was enhanced by nano silymarin into 37.5 and into 23.9% when treated with silymarin. An extreme high significant reduction was the result with EAC mice for GSH reduced and that was enhanced into 7.05 by nano size and into 10.9% with silymarin.
10- Finally the liver histological pattern for EAC bearing tumor showed congested portal vein and activated kupffer cell cytoplasmic vaculation of hepatocytes. Treatment with nano silymarin oral injection showed complete absence of fibrosis and silymarin treatment showed dilation for blood sinusoids in a normal shape.
