Abstract

Atotal number of 65 sample including The 65 Gram positive bacteria only focused on Staphylococcus aureous ,Corynebacterium psudotuberclosis for bacteriological examination . 65 samples 18 (27.69%) isolates of bacteria were recovered which categorized as 12(18.46%) C.pseudotuberclosis ,6(9.23%) S.aureous.
15 abscessiated prescapular l.n sheep origin, 2 abscessiated prefemoral l.n sheep origin, 3 abscessiated pulmonary abscess sheep origin, 3 abscessiated mesenteric l.n sheep origin, 11 abscessiated retropharyngeal l.n sheep origin, 6 abscessiated parotid l.n goat origin, 13 abscessiated submandibular l.n goat origin, 8 abscessiated hepatic abscess sheep origin , 3 abscessiated mammary l.n sheep origin, 1 abscessiated Subcutanus abscess sheep origin. were collected from private sheep and goats farms,butcher’s shop ,familes Matrouh homes , Ras ELhekma ,ELnasr Vilage ,ALsholahy Vilage and ELkaser .were examined during 12 month from November 2020 to November 2021 for examined anatomical distributed of subcutaneous and l.n abscesses with identification of clinical features and isolation of causative agents that cause these abscesses in sheep and goats in Matrouh Government.
For isolation of this bacteria the collected swabs for bacteriological examination ,Aloopful from pus sample was steaked on to nutrient agar,Mannitol Salt agar ,brain heart infusion agar and blood agar medium .
PCR was carried out on isolated S.aureous to detected enterotoxin gene in the six strains obtained . In PCR for enterotoxin gene detection, interesting all strains isolated from the different lymph nodes of sheep were positive for Sea enterotoxin except prescapular and submandibular lymph nodes.
ERIC-PCR was carried out on selected isolates of investigated 12 strain C.pseudotuberclosis to detect its genotyping.
ERIC-PCR results
Tewelve tested C.pseudotuberclosis investigated by ERIC-PCR analysis ,subtyped at to 4 ERIC-PCR types.
No 2,6,9,12 isolates respectively were clustered in same cluster (c1) with a dissimilarity of 7%,No8,10and 11 isolates were clustered together (c2) with a dissimilarity of 4% ,No 5 and 7 shown in the same cluster (c3) with adissimilarity of 1% ,and No 1,3and 4 isolates respectively were clustered in same group (c4) with adissimilarity of 7% ,the two cluster (c1and c2)differ from the other two cluster (c3 and c4) by about 25%
Positive ERIC-PCR 12 C.pseudotuberclosis strains susceptible to each of the following antibiotics E,DO,CIP,AT,CF,AMX,GEN,K,MUP,CD,AMP But strains were resistant to each of the following antibiotics:
CF,AMX,AMP,MUP,GEN.
Positive PCR 6 S.aureus strains was susceptible to each of the following antibiotic
DO,CIP,AT,GEN,K,CD,E
But strains were resistant to each of the following antibiotics PCR is a more efficient confirmatory tool for detection of these bacteria especially C.pseudotuberclosis and S.aureus
ERIC-PCR is one of the best methods to confirm identification , detection degree of similarity and dissimilarity between isolates in bacteria especially Corynebacterium pseudotuberclosis in addition to recognizing the source of infection and cross contamination.
Injecte the suspect microorganism in IP 20 male Guinea pigs then give result Staruass reaction ,inflammation in testes and orchites give confirmatory to its 12 strain C.pseudotuberclosis where ERIC-PCR relation ship reveals 4 subtypes.
In general the results of this study were particulary important to understand epidemiology,best antibiotics used in managing cases,control the C.pseudotuberculosis and/or S.aureus infection.
