Abstract

Background: Ischemic heart disease is a leading cause of death in human population and protection against ischemia-reperfusion (I/R) injury associated with myocardial infarction (MI) remains a challenge. So, cardio-protection against I/R injury should be considered. Geraniol is acyclic monoterpene with multiple pharmacological activities. However, its possible protective role in myocardial I/R Injury has not been elucidated. Aim: This study investigates the protective effect of Geraniol alone or in combination with Coenzyme Q10 in experimentally induced cardiac I/R injury. Method: Fifty rats were divided into five groups (n=10), Sham, I/R, geraniol and/or coenzyme Q10. Left anterior descending coronary artery was ligated for 30 minutes then released. The success of the model was assured by carrying out ECG before and all through the experiment. After reperfusion for 120 minutes, blood Samples were collected and the heart was excised detecting the infarct area and for further histopathological and biochemical analysis. Results: I/R injury group showed significant decrease in levels of SOD, Bcl2, GSK-3Ý and Ý-catenin as well as increased levels of TNF-Ü, Nf-KB, ICAM-1, Mpo, CK-MB, LDH, and cTn-1. While Pre-treated groups showed significant improvement of all biochemical parameters. These results were confirmed by histopathological examination as well as detecting the percent of infarct area of the area at risk. Conclusion: Geraniol or coQ10 significantly protected the heart via their antioxidant, anti-inflammatory and anti-apoptotic potential, while the combination almost normalized the biochemical levels and histopathological data. So, they can be considered as promising cardioprotective drugs
