Abstract The urgent need for new, efficient and safe antimicrobial agents has become a clear challenging target in drug discovery nowadays. Soil samples contain many microorganisms that produce antimicrobial agents of wide spectrum of activity. Therefore, in the present study twenty-eight soil samples were collected from different locations in Egypt during the period from October 2007 to October 2011. The different locations were Misr International University, Maadi, Marsa Matrouh, Nasr city, Al Ain El-Sokhna, Ismailia, Saint Catherine, Giza, and a field of cabbage in Dakahleya. from October 2007 to October 2010 Bacillus sp. were isolated from twenty-four soil samples but only nineteen soil samples contained Bacillus sp. active against MRSA or VRE. from October 2010 to October 2011 four more soil samples were collected for the isolation of Actinomycetes. Bacillus sp. and Actinomycetes are both spore forming bacterial strains found in soil that have activity against various microorganisms. Bacillus sp. isolates in this study were tested against resistant bacteria like Methicillin resistant Staphylococcus aureus (MRSA) and Vancomycin resistant Enterococcus sp. (VRE). Out of the hundred isolates, fifteen showed potent activity against MRSA but only two isolates Abstract 2 showed slight activity against VRE. Bacillus sp. encoded B7S5 was characterized morphologically, and biochemically as Bacillus subtilis. Eleven Actinomycetes were isolated, two promising Actinomycetes isolates encoded A1S19 and A10S19 showed potent antimicrobial activity against Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 29213, Candida albicans ATCC 10231.
