Abstract abilities of each of the two purified proteins were tested in a murine sepsis model of infection. Results: Eighty seven A. baumannii proteins were predicted to be localized in the outer membrane. Analysis of these proteins identified the DcaP like protein (DcaP) and the organic solvent tolerance protein (Ost) to contain the highest number of the best scoring T-helper cell epitopes. In order to ensure the global applicability of the proposed vaccine, the population coverage analysis indicated that both Dcap and Ost are predicted to associate with a large number of HLA-DR alleles that are common in 11 populations worldwide. They have an average coverage score of 89.03% and 81.27%, respectively, in all of the 11 tested populations scoring more than 90% coverage in 7 of those populations. Both proteins were successfully purified as His-tagged proteins. Animal testing demonstrated that Ost protein is immunogenic and induced a significant level of protection against a lethal challenge of A. baumannii whereas DcaP protein did not achieve such success. Conclusion: The data obtained in this study paves the road for the production of a novel vaccine against A. baumannii that would offer global protection. |
