Abstract

MicroRNAs (miRNAs), small non-coding endogenous RNA molecules with 18-25 nucleotides length, are implicated in regulating gene expression at the posttranscriptional level. Syndecan-1 (SDC-1), cell surface heparin sulphate proteoglycan acting, is known to regulate the expression of many miRNAs. Our study revealed that SDC-1 silencing using siRNA approach differentially regulated the expression of hsa-miR-182-5p, hsa-miR-186-5p, and hsa-miR19a3p, but had no effect on hsa-miR-222-3p expression in breast cancer cell lines Sum-149, MDA-MB-231, MCF-7. Using anti-miR-182-5p transfection, the enhanced cell proliferation and migration was partly inhibited associated with downregulation of RhoA mRNA expression in SDC-1 knockdown MCF-7 cells relative to controls. Overall, this study suggests that SDC-1 may regulate cell proliferation and migration via modulating hsa-miR-182-5p and Rho A expression
