Abstract

The aim of the current study was to investigate the effect of season, ovarian side, collection technique, follicle diameter and corpus luteum (CL) on yield, quality and in vitro maturation of Egyptian buffalo oocytes, also, enhancing the maturation rate by addition of different hormones and using of different maturation media. Ovaries were collected from abattoirs allover the year and cumulus oocytes complexes (COCs) were aspirated from visible follicles (2-6 mm). Ovaries were classified according to bearing CL into with and without CL. Oocytes quality were classified into excellent, good, fair and denuded. Only excellent and good quality oocytes were used for in vitro maturation medium supplemented with 10% FBS, 10 μg/ml ovine FSH, 10 IU/ml hCG, 1 μg/ml Estradiol and 1% antibiotic antimycotic. Results revealed that a significant difference between the four seasons with significant (P<0.5) increase of oocyte yield, quality and maturation for winter and spring than autumn and summer seasons. The left ovary was better for oocyte yield, quality and maturation. The slicing technique resulted in higher oocyte yield and quality than aspiration technique. Oocytes aspirated from medium follicles (3-6 mm) were of higher in quality and maturation rate. Recovery rate, quality and maturation rate of oocytes were higher for ovaries without CL than ovaries with CL. Maturation rate improved significantly (P<0.05) through addition of different hormones and TCM-199 is better for oocyte maturation and differs significantly (P<0.05) than Ham’s F-10 and DMEM .In conclusion, the winter season, left ovary, slicing technique, medium sized follicles and ovaries without CL showed positive effects on recovery rate, quality and in vitro maturation of buffalo oocytes and also, different hormone supplementation and using of TCM-199 enhanced oocyte maturation.
