Abstract SUMMARY γ- Ray is a type of ionizing radiation that is used in many medical and industrial applications. The present study was carried out to evaluate the genotoxic effect of γ-ray in adult male mice by using cytogenetic techniques, gel electrophoresis and colorimetric methods to evaluate the protective role of spirulina against induced damage. Spirulina is blue green algae that administrated intraperitoneally in two different doses day after day for two weeks alone or before 4 Gy of γ-irradiation exposure to evaluate its possible protective role against damage induced by ionizing radiation. Chromosomal aberration of metaphases from mice bone marrow, gel electrophoresis for determination of DNA fragmentation of experimental mice spleenocytes, sperm morphological anomalies and colometric method for serum lipid profile measurement, all previous mentioned techniques were used in that study, and showed the following: • γ- ray was found to increase the frequencies of both structural and numerical chromosomal aberration significantly when compared with control animal mean values and animals received spirulina doses of 200 mg/kg. bw. and 400 mg/kg. bw. whereas animals received spirulina 200 mg/kg. bw. and 400 mg/kg. bw. then exposed to 4Gy dose of γ-irradiation showed obvious amelioration in both structural and numerical aberrations compared to irradiated animals. • DNA fragmentation in spleenocytes was performed by gel electrophoresis technique, black and white photograph showed clear cut damage to DNA of animal exposed to 4 Gy of γ-ray, and visibly improvement in tracks of animals treated with spirulina in both doses before irradiation. • Animals treated with spirulina clearly showed an enhancement in sperm morphology that was revealed in the values of mutation factors and mutation index of head and tail sperm morphology, and there were clear reduction in these values in mice groups received spirulina before irradiation. • The present study revealed that spirulina has great efficacy in the reduction of total cholesterol level, triglyceride and noticeably amelioration in serum LDL/HDL ratio in animals treated with spirulina alone and in animals treated with spirulina then exposed to radiation. • Finally, spirulina has ability in body weight reduction, especially in animal groups received high dose of spirulina (400 mg/kg.bw.). |
