Abstract Objectives: This study was conducted to evaluate the effects of topical application of hyaluronic acid on oral ulcer of rats receiving methotrexate. Material and methods: Forty five male albino rats wgt. 200-250 were selected in this study and divided into 3 groups (15 rats each): group I: The rats received sham treatment and considered as a control, group II: The rats received MTX. (10mg/kg/week), group III: The rats received MTX (10mg/kg/week) and hyaluronic acid (240mg/100gm twice daily) treatments continued till sacrifaction date. Ulcer was inducted after one week of methotrexate injection in groups (II and III). Ulcers were done at every animal’s cheek by using biopsy punch. According to date of sacrifaction each group divided into three equal sub groups at 4th day, 7th day, and 14th day; right sides cheek specimens were removed and immediately prepared for paraffin sections and stained with H&E stain left sides of cheek specimen were put in glutaraldehyde fixative solution for electron microscopy examination. Results: At 7th day post ulceration there were a significant decrease in inflammatory cells infilteration in group III, also there were moderate widening of intercellular spaces, less destructed desmosomal junctions and more packed collagen fibers. While in group II there were significant increase in inflammatory cells percent area, no regaining of epithelial cells to their attachement with destructed basal lamina. Conclusion: Based on the previous results we concluded that topically application of hyaluronic acid on oral ulcer on rats immunosuppressed by methotrexate lead to faster healing of the oral ulcer. |
