Abstract This work a1ms to clarify the induction of the ”Crabtree effect” in liver cells through measuring certain biochemical and histochemical parameters in livers of Schistosoma mansoni infected mice. The liver is one of most important targets for toxic substances. At the same time it represents the main site for detoxication in the orgamsm. from the biochemical point of view, the knowledge about this organ is much more advanced than that regarding other tissues. The curative effect of oil extract of Curcuma Jonga were tested and compared to praziquantel as the effective drug against all schistosome species occurring in man. Measurment of protein, glycogen, glucose, alkaline and acid phosphatases, glucose-6- phosphatase, AMP deaminase, adenosine deaminase, urea concentration, pyruvate kinase (PK), phosphoenol pyruvate carboxykinase (PEPCK), and PK/PEPCK ratio were performed. In addition, to measurments of body weight, liver weight, liver weight/ body weight in normal and S mansoni infected mice will be investigated. Moreover worm burden and ova count in mice infected with S mansoniwere elucidated. Abstract Mice were given a single injection with S. mansoni through subcutaneous infection with 50 cercariae followed by a praziquantel treatment (500 mg/kg body weight) after one month, two months on two successive days. Others were treated with Curcuma longa, 1st month post infection for two months (300 mg/kg body weight) twice a week. Also, control groups were treated either PZQ or C longa extract. It was interested in the present work to study the effect of the medicinal plant C longa on the enhancement of the liver studied metabolic parameters. C Jonga enhanced the concentration of protein, glycogen and glucose by elevating them after being reduced by infection while PZQ- treatment revealed more reduction. Alkaline phosphatase level was increased with infection. Then C Jonga treated mice showed a marked reduction more than PZQ- treatment. C Jonga induced normalization in PK level, which was elevated in infected mice while PZQ- treatment induced more elevation of this enzyme. A relative improvement m AMP- deaminase and urea concentration occurred in response to C longa than to PZQ treatment. The present study showed that due to Abstract schistosome infection, body weight was reduced while liver weight increased. C longa caused enhanced in body weight while PZQ has no effect. The formation of granuloma around schistosome eggs in the liver produced inflammation. C longa extract and PZQ were effective in reducing worm burden, ova count and granuloma size in infected - treated m1ce. C longa proved to be highly effective against S mansoni. |
