Abstract

Considering the several theories and suggestions enlightening the relevance and design of novel compounds that are used for the treatment of cancer, encouraged by a comprehensive literature survey focusing on bendamustine, dovitinib and Hoechst 33258 as antitumor drug containing benzimidazole moiety in its structures, the synthesis of novel compounds that carry the main structural features was promoted to attain a curative effect towards cancer. In order to obtain the target compounds, different synthetic routes were adopted and found to be successful. The present investigation necessitated the synthesis of five unavailable intermediates, guided with the published literatures. In the present study, forty six novel compounds that are not mentioned in the available literature were synthesized. The structures of these compounds were elucidated through elemental microanalyses, IR, 1H-NMR, 13C-NMR and mass spectroscopy.The newly synthesized compounds were subjected to preliminary biological screening for their antitumor activity. Fourteen compounds are active as showed % surviving of tumor cells is less than 30%. IC50 for six active compounds was determined in vitro against MCF-7 tumor cell line using doxorubicin as a reference antitumor agent. Two compounds 133e and 134c showed moderate activity against MCF-7 cell line while four compounds 106a, 133b, 133d and 142h showed low activity.
