Abstract Cholesterol is an essential component of cell membrane and having great important precursor for vitamin D synthesis, bile acids, and various steroidal hormones. Cholesterol is excreted from the liver in bile which aids in the digestion of fats. There are many diseases can be caused by diabetes. If not under controlled. Diabetes can cause blindness if not treated. Diabetes is leading to cause kidney failure. High blood sugar can harm nerve including peripheral diabetic neuropathy. Present study aimed mainly: 1- Clarify the adverse effect of hypercholesterolemia and diabetes and focus on the effect of two hypocholesterolemic agents (from different groups mainly (atorvastatin) and (fenofibrate) drugs on many metabolic changes. 2- Histopathological examination for selected tissues which are liver and testis additionally aorta from experimental rats to configure the effect drugs and correlation with results. 3- To focus on these biochemical measurement and if there any a fundamental disagreement between the drugs that used depending on the mechanism of action. 4- To fulfill such objectives the following parameters were selected to be studied include vitamin D synthesis, bile acids, certain steroidal hormones (cortisol, testosterone, aldosterone) and lipid profile(TC, HDL-c, LDL-c, serum glucose, fructosamine and TAG) in experimental hypercholesterolemic diabetic rats. Experimental design One week after acclimatization experimental rats received a diet enriched with 2% cholesterol, 2% saturated fat (butter fats) and 0,5% bile acids (cholic acid) supplemented with 0.01% thiouracil in drinking water for 12 weeks to induce hypercholesterolemia. Hypercholesterolemic rats rendered diabetic by administration of single dose intraperitoneally (90mg/kg bodyweight) of alloxan. Diabetic hypercholesterolemic rats were further categorized into Three groups (n=25) as follow group (1): Diabetic hypercholesterolemic group (Control) (DH) group (2): Atorvastatin group (A). Diabetic hypercholesterolemic rats kept on cholesterol enriched diet, received atorvastatin (10mg/kg/day) for 4 and 8 weeks. group (3): Fenofibrate group (F). Diabetic hypercholesterolemic rats kept on cholesterol enriched diet, received fenofibrate (100 mg/kg/day) for 4 and 8 weeks. Another group received no drugs, kept on normal diet was added as normal control group. |
