Abstract

Chemotherapy treatments cause dry mouth by damaging the salivary glands. Patients with severely dysfunctional salivary glands often suffer from impaired speech, infection, and loss of taste perception. The development of a non-toxic selective cytoprotective agent that preferentially protects normal tissues from chemotherapy toxicity, without protecting malignant tissues, is a major challenge in cancer chemotherapy research. Wheat germ oil possesses various biological properties as an anticancer and antioxidant agent. Seventy two adult male rats were selected for this study. They were divided into three groups, Group I (control) rats were injected intraperitonially with normal saline (n=12), Group II cisplatin injected intraperitoneally once a week for four weeks in a dose of (5 mg\kg) (n=30) and Group III cisplatin injection with the same dose, with daily administration of wheat germ oil in a dose of 1 ml\kg for 6 weeks (n=30). Two rats from group 1 and five rats from group II, III were sacrificed weekly; the parotid and sublingual glands were removed and prepared for (H & E) stain, immunohistochemical staining of PCNA and transmission electron microscopy study. .Results: serous acini of group II showed numerous changes more than mucous acini as loss of acinar outlines with hydropic changes and numerous of intracytoplasmic vacuoles more than group (III). The PCNA index showed decreased number of proliferative cells especially in group (II). The ultrastructural results, group (II) showed sever damage of the nucleus, dilated rough endoplasmic reticulum, large number of vacuoles and heterogeneous secretory granules. But group (III) showed less destructive changes. Conclusions: wheat germ oil protect the salivary gland from cytotoxic effect of cisplatin.
