Abstract

This study was carried out on 1281 Friesian dairy cows in private dairy farms and small stock holders located in Port-Said, Qaliubiya, Damietta, Monofia and those located
on the Cairo-Alexandria Desert Road to study the mastitis problem with special reference was done to Staphylococcal mastitis. Clinical examination, CMT and SCC, PCR and gene typing in addition to sensitivity against some antibiotics were done in this study. 91 cows with 335 QMS were examined bacteriologically. They recorded that 122 QMS were positive while 231 QMS were negative for bacteriological examination. 97 strains of S. aureus, 17 strains of CNS and eight strains of streptococci isolated from positive QMS. PCR was used for direct detection of S. aureus in raw milk as a rapid diagnostic tool. Genotyping of virulence factors encoding genes of S. aureus was done. Coagulase gene, Thermonuclease gene, gene segment encoding the immunoglobulin G binding region and the Staphylococcal enterotoxin genes group (SEs) {A, C, D, G, H, I, J and L} categorizing the pathogenic virulent S. aureus into enterotoxigenic and nonenterotoxigenic. Sensitivity test against some antibiotics was done for both S. aureus and CNS. S. aureus was sensitive to Gentamycin and Amoxicillin /Calvulanic acid, while CNS strains were sensitive to Amoxicillin /Calvulanic acid and Cloxacillin.
