Abstract

Tetanus toxin was prepared and tested for sterility and safety. Also MLD and Lf has been done to know the potency of the toxin. Inactivation of the toxin has been done to convert it into toxoid then adjuvants (aluminum hydroxide gel, PLGA 50:50 and PLGA (80:15+50:50) were added to toxoid. The prepared vaccines injected in Guinea pigs and rabbits (each vaccine 10 G. pigs and 10 rabbits) and samples collected to detect the antibody titer. The results showed that sera of rabbits and Guinea pigs contained tetanus vaccine with microparticles are higher in antibody titer and also have prolonged protective period. The antibody titer resulting from PLGA (85:15 and 50:50) was higher than that of PLGA (50:50) only. The duration of immunity has been prolonged in PLGA (85:15 and 50:50) than that of PLGA (50:50) only. Measurement of antibody titer by serum neutralization test and ELISA confirmed these results
