Abstract Many cellular functions are regulated by changes in gene expression. Thus, quantification of transcription levels of genes plays a central role in the understanding of gene function and of abnormal alterations in regulation that may result in a disease state. There is resurgence in the use of herbal medicines worldwide. In contrast to chemical drugs, herbs have sometimes been claimed to be non-toxic, because of their natural origin and long-term use as folk medicines. Using of thymol in form of adjuvant induce the immune response and cytokines production. Therefore, the outcome of this work was to demonstrate the feasibility of using some physical treatments on MDR E. coli 026 and follow up the gene expression of some proin:flammatory cytokines and toll-like receptors-4 ex-vivo. In the present work, The susceptibility pattern of E. coli 026 revealed that it was sensitive to cefoperazone (CFP), cefuroxime (CXM) , ciprofloxacin (CIP) ,gentarnicin(CN), irnipenem (lpm) and amikacin (AK) and while it showed resistant to more than three antibiotics Nitrofurantoin (F), Streptomycin (S), Tetracycline (TB), Trimethoprime/sulphamethoxazole (SXT) and Ampicillin /Sulbctarn (SAM), hence meeting the criteria for multidrug resistant. Regarding the calculation of Colony forming units (CFU) of E. coli 026 strain, it was determined and the bacterial cell count was 36x IOII/rn!. Concerning the inoculation of rabbits with MDR E. coli 026 strain in different physical treatments within different time points, marked gene expressions of TLR-4 and some proin:flammatory cytokines mRNA was revealed. |
