Abstract The present investigation aimed at assessing the utflity of otato anther cuture technique for the in vitro selection in otato plants tolerant to salinity and lines resistant to Fusar lam. Three dihaploid (2X = 24 chromosomes) populations (A, B and C) of potato (Solanum tuberosum L) were used as anther donor plants. A) In vitro selection for sodium chloride tolerance: In vitro seection for sodium chloride tolerance had been made using microspores (anthers) and androgenic derived embryolds as follow: I-Tn vitro selection for microspore tolerance: Anthers were cultured in liquid medium containing 0.0 and 0.4% sodium chloride. The selected embryoids from sodium chloride containing rneum were devide into two groups and regenerated as follow: — Short term selection: the first group of selected embrycids was regenerated on salt—free medium. Long term s&Iection: the second group regenerated on medium containing 0.4% NaCl (long term selection). II-In vitro selection for ernbryoids tolerance: — In the single step selection, androgenic derived ernbryoids were cultured on medium containing 0.0%, 0.4% and 0.6% Nacl. - Step—wise selection was also used by culturing androgenic derived embryolds on medium supplemented with 0.2% NaC1 and ait concentration was increased step—wise by 0.2% up to 0.6% within two weeks. |
