Abstract In the present study 600 fertile SPF egg samples representing 30 batches used in the Central Laboratory for Evaluation of Veterinary Biologics (CLVB) were collected for evaluation. Different materials were sampled from the eggs including (egg yolk, egg albumin, allantoic fluid ) and used in screening of any possible virus transmission and/or antibodies against some selected viruses: The currently used quality control protocols applied in the CLVB are : (ELISA test for the presence of CAV and REV antibodies as a contaminant in egg yolk samples, ELISA test for the presence of ALV antigen as a contaminant in egg albumin samples, AGDT for the presence of IBD, ILT, POX, Reo virus antibodies as contaminants in egg yolk samples, HA test for the presence of Influenza virus type A, NDV, EDS76, avian Paramyxovirus type 2 antigens as contaminants in allantoic fluid) revealed the negativity of all SPF egg samples for both antigens and antibodies. An improved quality control protocol for evaluation of SPF eggs was applied. The improved protocol depend on the use of PCR to detect the vertically transmitted viruses (CAV, REV, and ALV) which represent the main causes of the problems in SPF projects. Application of PCR as an added value to the currently applied quality control protocol in testing the 30 egg batches revealed their freedom of the vertically transmitted viruses (CAV, REV, and ALV). This study proposes the importance of PCR as an improved protocol in detection of vertically transmitted viruses (CAV,REV, and ALV) as a confirmatory assay. |
