Abstract

Stem or progenitor cells are defined by their capacity for cell renewal and asymmetric cell division, which leads to their differentiation and formation of one or more mature tissues. Stem cell therapy holds great promise for the repair of injured tissues and organs, including the kidney. The aim of the present investigation was to demonstrate the effect of transplanted haematopoietic stem cells on renal functions in experimental nephropathy in rats. Thirty adult male albino rats, weighing 150-200 gram, were used in this work. The rats were classified into three groups Normal control group Nephropathy non-treated: in which rats were made nephropathic by single intravenous injection of adriamycin in a dose of 5mg/kg Nephropathy stem cell - treated group: in which nephropathic rats were treated with single intravenous injection of stem cells At the end of the experiment in each of the previously mentioned groups 24 hrs urine samples were collected by metabolic cages to estimate urine volume, urinary protein, and urinary creatinine. Retro orbital blood samples were used to estimate serum urea, serum creatinine and serum albumin. Creatinine clearance was calculated. The rats were sacrificed and kidneys were excised for histopathological and immunohistochemical studies.The results showed that induction of nephropathy by adriamycin was associated with significant increase of serum urea, serum creatinine and 24 hrs urinary protein. Also there were significant decrease in creatinine clearance and serum albumin when compared to the corresponding values in normal control group. Histopathological examination revealed significant increase of the percentages of affection cloudy swelling, hyDROPic degeneration & necrosis) of PCT, DCT and CT, Also H&E stained sections from the kidneys of nephropathy nontreated group revealed focal tubular necrosis and the renal corpuscles showed varying degrees of degeneration. PAS-stained sections of nephropathy non-treated group showed thickened basement membranes in Bowman’s capsule, glomerular capillaries and some renal tubules. In masson’s trichrome stained sections of nephropathy non treated group an increase in the green stained collagen fibers was observed in the interstitium, in the area of focal tubular necrosis as well as in the mesangium. Immunohistochemical stained sections, from nephropathy non- treated group showed negative staining for the monoclonal mouse Anti-human CD 45.
