Abstract

This study is an experimental study and was designed to investigate the possibility of HUCB CD34+ cells to home in the pancreas and kidney of diabetic mice.
This study included three groups: group (1) was treated by HUCB cells after induction of diabetic, group (2) was the diabetic group and received no treatment, group (3) was the control group.
Isolation of HUCB CD34+ cells was carried out in Stem Cell Unit in the Physiology Department, Faculty of Medicine, Suez Canal University after cord blood collection in the Gynecology and Obstetric department, Faculty of Medicine, Suez Canal University
Induction of diabetes was done by STZ through a single intraperitoneal injection. HUCB CD34+ cells were injected intravenously in the tail vein. Blood glucose level was measured throughout the study. After three weeks all animals were sacrificed, pancreas and kidney tissues were frozen and were assayed for the presence of human DNA (human ALU sequence) using real time PCR. The collected data was statistically analyzed using SPSS and MedCalc statistics softwares.
Blood glucose level was elevated in group (1) and (2) which was injected by STZ. After treatment with HUCB CD34+, the blood glucose level in group (1) decreased significantly. PCR assays showed that HUCB CD 34+ cells home in the pancreas and kidney tissues of group (1) only which was treated by the stem cells and consequently improves diabetes.
