Abstract The objective of this study was conducted to search for some new natural antibiotics against broad spectrum microorganisms and to determine the probable mechanism through which the new antibiotic exerts its action on test organism. from screening program, the antibiotic extracted from Streptomyces CAS 10 exerting a pronounced antimicrobial activity against Gram-positive and Gram-negative bacteria. Results of this study dearly indicated that, the antibiotic has no obvious effect on cell wall synthesis and the target for its action is the nuclear area where it strongly curtailed the synthesis of Staphylococcus aureus soluble phosphorous, RNA, DNA besides a cytoplasmic pool target, protein synthesis. The LD50 of CAScin in mice was found to be 28.7 mg/kg body weight. CAScin has nephrotoxic, hepatotoxic where it decreases total proteins and albumins. In the mean time it increases in urea, uric acid, globulin, and liver enzymes. also it increase the risk of atherosclerosis as it increases total cholesterol, LDL-cholesterol, triacylglycerol CAScin showed remarkable cytotoxic activity in vitro against Colon carcinoma cell line (HCT116) & Breast carcinoma cell line (MCF7) at different concentrations. |
