Abstract
This work was carried out in the Plant Tissue Culture Laboratory at El-Sheik Zuwyed Research Station - Desert Research Center (DRC) North Sinai – Egypt, during the period from 2004 to 2006.The objectives of this study were to investigate the preservation of germplasm of Ecballium elaterium and Capparis spinosa using tissue culture technique and study their response to osmotic inhibitor, low temperature and dark conditions on callus and plantlet preservation. These results showed that the best preservation condition of Ecballium elaterium at temperature 4°C, in dark and the modify MS medium with sucrose concentration was 60g/l for six months. Shoot length 3.60 (cm) and leaves number/ shootlet (5.67). Callus fresh weight of Ecballium elaterium was preserved on 45g/l sucrose (3.04g). Also, modify MS medium at 22°C. in light for five months. Existed the best condition of Capparis spinosa on 45g/l sucrose modify MS medium at 22°C. in light for three months, shoot length 0.67 (cm), leaves number/ shootlet (52.00), shoots number/ explant were (8.67).
