Abstract

Thermophilic bacteria thrive at temperatures 65-85oC. In recent years much interest has been focused on the characterization of these microorganisms, and in particular on their enzymes. Thermophilic lipases are currently attracting enormous attention because of their biotechnological potential. It was thus aimed in the present study to isolate a local thermophilic bacterium capable of producing lipases/esterases. Evaluation of culture conditions affecting enzyme activity was also investigated. The main goal was to clone and express the genes and to purify and characterize the produced enzymes. To achieve the goal, the experimental work was conducted in four parts and the results obtained are summarized as follows. 1-In the first part, enrichment of soil samples led to the isolation of 42 isolates that grew well at 60oC and showed good lipolytic activities.
