Abstract

The improvement of fertility of frozen thawed of goat and sheep spermatozoa is becoming
very important. Therefore, the study of suitable and available method of freezing under
developing countries conditions is very necessary. Moreover, the studies of the effect of different
levels of glycerol and ethylene glycol compared to the control which had no concentration of
glycerol or ethylene glycol, the effect of freezing processing on the content of intracellular
concentration of ATP of goat spermatozoa, and effect of dehydration medium of extension
compared to routine extender on the conception rate of ewes could be considered as a good
evaluation of the new technique of freezing and dehydration.
This study was carried out at Faculty of Agriculture, Suez Canal University, Ismailia,
Egypt and Animal Breeding and Husbandry Institute, Gottingen university, Gottingen, Germany
under the channel system between Egypt and Germany ( 1994-1997).
Therefore, the present study was carried out on the following:
Part 1: a) Semen producing capacity of rams by using partial depletion (3 Ejaculates/day
for 10 days).
b) Effect of successive ejaculates, age of ram, and incubation time on motility, fructose
concentration wd fructose utilization of sheep spermatozoa
c) Effect of cryopreservation as pellets, on the cold surface of paraffin wax on the
acrosome integrity of sheep spermatozoa.
Part 2: Comparative studies between the different methods of freezing goat spermatozoa
(straws & dry ice pellets) and the new method of freezing as pellets on the cold surface of
paraffin wax.
Part 3: a) Comparative study between sucrose as a cryoprotectant and glycerol as a
standard cryoprotectant for freezing goat buck semen.
b) Dehydration process and its relation to cryopreservation of goat spermatozoa.
c) Adenosine Triphosphate (ATP) of spermatozoa as a new predicting method of fertilizing
ability of goat buck spermatozoa.
